IPEC Europe Le monde l’excipient : excipient specificity
IPEC EuropeIPEC Europe, the International Pharmaceutical Excipients Council Europe, is an association which serves the interests of producers, distributors and users of pharmaceutical excipients.
Together with its sister associations IPEC Americas and IPEC Japan (JPEC) the Council is member of TriPEC whose global membership extends to more than 200 companies.
IPEC Europe represents the views of its members to appropriate regulatory bodies (European Commission, EMEA, European Pharmacopoeia) and is recognized by government agencies around the world as the voice of European producers, distributors and users of pharmaceutical excipients.
For this it develops position papers and it contributes to global harmonization efforts in the fields of regulations, regulatory standards and pharmacopoeial monographs for excipients.

Natural, synthetic, biological…Industries belonging to multiple sectors Commodity ingredient Dedicated business or better dedicated manufacturing siteUsing quality management standards
Contrary to APIs, excipients are usually not specifically made for use in medicinal products:Examples :
•Cellulose production 250 million t/y Cellulose for pharma 0.05 million t/y
•Propellants / Refrigerants:HFA 134a (norflurane):98% for cooling / 2% for pharma
Excipients = API?
Average of 1-5% estimated for the pharmaceutical industry (mainly food, food additive, cosmetic ingredients…)High volume, low valueLarge spectrum of companies (small to large)Multiple sources and complex distribution networkRegulated by the main industry served , voluntary quality systems, quality management cultureContinuous process, bulk processNo dedicated expertise in pharmaceutical marketPer regulation, novel excipients and excipients used as API, are API
Differences entre Excipients et API
Active Ingredient ExcipientManufacturing plans dedicated to pharmacy
Multiple usage away from pharmaceutical industry
High value, small volume High volume, low value
Limited source of products Multiple sources
More regulated Less regulated, voluntary systems
Clear rules imposed Customer-oriented food/cosmetics/general market
GMP practices Quality management culture
Process/batch system Continuous process, bulk process
More experts in regulatory affairs in pharmaceutical world

• Directive 2004/27/EC (art 46f)• This point shall also be applicable to certain
excipients, the list of which as well as the specific conditions of application shall be established by a Directive adopted by the Commission in accordance with the procedure referred to in Article 121(2).

Key Deliverables of the Project:International in scopeBased on the IPEC-PQG GMP Guide 2006Based on IPEC GDP Guide 2006Aligned to ISO 9001 (2000 and 2008 editions)Many Excipient suppliers are already ISO certified
Key Project Principles:Evolve existing best practices from all known sectorsClassify ExcipientsMust include GDP and GMPAuditor training and competency requirements must be defined The scheme must apply to the majority of the existing Excipient industry without raising the bar too high

Conclusion
Trading environment is becoming more complex (global market, distribution and traders, internet supplies, sub-standards products..)Regulatory requirements for excipients are non consistent and non harmonisedRegulations applicable to API are impacting affecting excipients despite of its non relevance in most of the casesRisk mitigation tools for excipients
